Biologically, the peptide is studied mainly in the context of immune cell development and regulation. It is produced in the thymus and in several other tissues, and it appears to influence the maturation and activity of T cells and other immune populations. Laboratory work describes effects on cytokine production, on the balance between T cell subsets, and on the function of dendritic cells. Much of this evidence comes from cell culture and animal models, so the extent to which the same pathways operate in humans remains an open question.
Clinical interest has centered on chronic viral hepatitis, on immune restoration in various conditions, and on use as an adjuvant intended to improve responses to vaccines. Trials have reported mixed results, and regulatory status differs sharply between countries; in some places it is a prescription product, while elsewhere it is sold without an approved therapeutic indication. Because published studies vary widely in design, population, and endpoints, comparisons across them are difficult and no single conclusion covers the whole literature.
Thymosin alpha 1 is a short peptide of 28 amino acid residues that derives from the amino terminal region of a larger precursor protein known as prothymosin alpha. The peptide carries an acetyl group on its first residue and contains no disulfide bonds or carbohydrate chains. Its sequence is highly conserved across mammalian species, which is one reason laboratories treat it as a molecule with a defined and reproducible structure rather than a variable tissue extract. The name follows an early naming convention for thymus-derived fractions and does not imply that the peptide acts as a hormone in the classical endocrine sense.

Immunological studies connect the peptide to multiple parts of the immune response. It has been reported to engage Toll-like receptor signaling, to promote dendritic cell maturation, and to influence the balance of T helper cell subsets. Changes in natural killer cell activity and in cytokine release appear in cell culture and animal models. These observations describe broad immunomodulatory behavior rather than a single defined receptor target, and the primary molecular interaction has not been settled.
Thymosin alpha-1 is a synthetic 28-amino-acid peptide whose sequence was first identified in extracts of bovine thymus tissue during the 1970s. The chain carries an acetyl group on its N-terminal serine. Its acidic residue content is high, which produces strong water solubility and an isoelectric point well below neutrality. Material supplied for laboratory and clinical use is manufactured by solid-phase peptide synthesis rather than purified from animal tissue. Different salt forms, such as the acetate, alter the counter-ion content without changing the peptide backbone.
Immune signaling studies link thymosin alpha 1 to Toll-like receptor pathways, particularly TLR2 and TLR9, on dendritic cells and other antigen-presenting cells. Activation of these receptors promotes maturation of T cells and increases natural killer cell activity. The peptide shifts cytokine output toward a T helper 1 profile, raising interferon gamma and interleukin 2 while modulating interleukin 10. Whether these effects translate into clinical benefit for any specific disease remains a subject of debate. Reported outcomes vary across trials and populations.
Thymosin alpha 1 is approved as a medicine in several countries, including Italy and China, for indications such as chronic hepatitis B and as an immune adjuvant. It is not approved by the United States Food and Drug Administration as a therapeutic product. In research settings the peptide appears in studies of sepsis, vaccine response, and oncology support, often with mixed or inconclusive results. The evidence base is uneven, and reviews note that many trials were small. Regulatory status therefore differs widely between jurisdictions.
The peptide was described in the 1970s as a component of thymic extracts, and early research focused on restoring immune function in immunodeficiency states. A synthetic version entered clinical development in the 1980s and is approved as a drug in several countries for conditions such as chronic hepatitis B and certain immunodeficiencies. Approval status varies widely by jurisdiction, and in the United States it is not an approved therapeutic. Regulatory and clinical positions differ, so statements about efficacy should be tied to specific indications and studies.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids, corresponding to the N-terminal fragment of prothymosin alpha. Its sequence begins with acetylation at the N-terminus, a modification that affects stability and receptor interaction. The peptide is acidic, with a calculated isoelectric point near 4.2, and carries no disulfide bonds, so its secondary structure is largely flexible in solution. Molecular mass is approximately 3108 daltons. The native form was first isolated from bovine thymus tissue, while pharmaceutical material is produced by solid-phase peptide synthesis.
Early work on thymic extracts in the 1960s described a heat-stable acidic fraction containing many polypeptides. Separation of that mixture yielded individual components, and thymosin alpha-1 was named as one of them on the basis of assays for T-cell activity. The first preparations came from calf thymus, while subsequent research and clinical material has been chemically synthesized. Nomenclature in older papers is inconsistent, and the same peptide sometimes appears under different designations, which complicates literature searches.
Most published studies on thymosin alpha-1 report changes in immune measurements rather than clinical outcomes, and findings differ across designs and populations. Whether the peptide signals through one defined receptor or through several less specific interactions remains an open question. Its reported circulation half-life of a few hours complicates comparison of dosing schedules across trials. Mechanistic claims are frequently drawn from isolated cell cultures, and how far those results extend to whole organisms is unresolved.
The second most common form of SCID after X-SCID is caused by a defective enzyme, adenosine deaminase (ADA), necessary for the breakdown of purines. Lack of ADA causes accumulation of dATP. This metabolite will inhibit the activity of ribonucleotide reductase, the enzyme that reduces ribonucleotides to generate deoxyribonucleotides. The effectiveness of the immune system depends upon lymphocyte proliferation and hence dNTP synthesis. Without functional ribonucleotide reductase, lymphocyte proliferation is inhibited and the immune system is compromised.
== Chestnut mimics == Bay horses also have reddish coats, but they have a black mane, tail, legs and other point coloration. The presence of true black points, even if obscured by white markings, means that a horse is not chestnut. Seal brown or dark bay horses are not chestnut but may be confused with a liver chestnut. Those unfamiliar with horse coat color terminology often call most horses "brown". including chestnuts. Brown, which may be difficult to distinguish visually from dark bay, is always accompanied by black points. Liver chestnuts, in particular, are mistakenly called brown or "seal brown". Silver bay horses typically have chocolate- to red-brown bodies with silvered mane, tail, and legs. The flat reddish-brown color and lack of easily identified black points can confuse even knowledgeable horse persons. Silver dapple horses usually hint at black or dark gray pigment at the roots of the mane and tail, and where their silver points end on the legs. Silvers look a bit "off"-chestnut. To further confuse matters, some flaxen chestnuts have silverish streaks in their manes and tails. However, genetic testing can clarify matters.

One-place studies of urban parishes are less common, since urban populations were migratory and analysis is more difficult when few of the families remain present for the whole period under study.
Type 1: Antigenically Ran2+, GFAP+, FGFR3+, A2B5−, thus resembling the "type 1 astrocyte" of the postnatal day 7 rat optic nerve. These can arise from the tripotential glial restricted precursor cells (GRP), but not from the bipotential O2A/OPC (oligodendrocyte, type 2 astrocyte precursor, also called Oligodendrocyte progenitor cell) cells. Type 2: Antigenically A2B5+, GFAP+, FGFR3−, Ran 2−. These cells can develop in vitro from the either tripotential GRP (probably via O2A stage), from bipotential O2A cells, or in vivo when these progenitor cells are transplanted into lesion sites (but probably not in normal development, at least not in the rat optic nerve). Type 2 astrocytes are the major astrocytic component in postnatal optic nerve cultures that are generated by O2A cells grown in the presence of fetal calf serum but are not thought to exist in vivo. Some researchers think bipotential O2A cells in turn have been derived from the GRP.

== Structure == The activin and inhibin protein complexes are both dimeric in structure, and, in each complex, the two monomers are linked to one another by a single disulfide bond. In addition, both complexes are derived from the same family of related genes and proteins but differ in their subunit composition. Below is a list of the most common inhibin and activin complexes and their subunit composition:
Ablukast (INN) is an experimental drug that is a leukotriene antagonist. It was investigated for potential applications in the treatment of inflammatory conditions, including asthma, skin disorders, and inflammatory bowel disease. It reached Phase III clinical trials, but development was discontinued in 1996.
Second Cup was founded in 1975 by Tom Culligan and Frank O'Dea in Toronto, Ontario. Culligan eventually purchased O'Dea's shares and expanded Second Cup into a 150-store chain. He sold it in 1988 to the founder of mmmuffins, Michael Bregman. As chairman and CEO, Khalil Al Gawad took Second Cup public in 1993. Between 1993 and 2002, it was owned by several American companies, including Coffee Plantation, Coffee People, and Caribou Coffee. Al Gawad sold Second Cup to Cara Operations Limited in 2002. In November 2006, Cara sold Second Cup to Dinecorp Hospitality, controlled by former Cara CEO Gabe Tsampalieros, who became chairman of Second Cup. Tsampalieros died on March 11, 2009. The trademark rights were subsequently split between Canada (the Second Cup Ltd.) and international (the Second Cup Coffee Company Inc.). Stacey Mowbray was head of the Canadian company, and Jim Ragas led the international side. Second Cup was featured in an episode of Undercover Boss that aired in March 2012 on the W Network. Under Mowbray's direction, Second Cup was presented as "a company that cares". In September 2012, the company launched Tassimo beverages. In April 2015, Second Cup started a rewards program, which allows users to earn points using a mobile app. Alix Box was the CEO and president at Second Cup Ltd. from 2014 until her sudden departure, in May 2017; she was temporarily replaced by Garry MacDonald. On April 12, 2018, Bregman announced an agreement with National Access Cannabis to develop and convert a network of recreational cannabis stores.
==== Notes ==== a See Freeze brand § Freeze branding as a painless alternative to mulesing. b See the subsection on human cryobranding in Freeze brand § Usage. c Irwin reports that keeping the iron stationary when branding a dolphin's dorsal fin, rather than rocking it as with livestock, produced a markedly clearer result. d Although adequate marks appeared on the elephant seal's skin at the time of cryobranding none of the marks produced lasted more than 1 year. e Down feathers grew white after cryobranding but did not persist in adult Mallards nor affect the outer contour feathers when they grew in. According to Greenwood, citing Foulks: "melanocytes continually arise from undifferentiated melanoblasts in the dermis during feather growth. Apparently the presence of whitetipped down resulted from the destruction of melanocytes associated with the tip of the developing feather, which were later replaced with active melanocytes before the deposition process terminated. Although birds were treated at ages of 5–10 days, and at various early stages of feather development, no apparent effect on pigmentation of contour feathers occurred." f Herpetologist Charles F. Smith described freeze branding as "[in]effective for bufonids [toads] and other taxa with granular skin surfaces." g All salamanders tested were seriously injured, and some later died. The author does not recommend his method be copied.
== Synthesis and Post-translational Modification == Proinsulin is synthesized on membrane associated ribosomes found on the rough endoplasmic reticulum, where it is folded and its disulfide bonds are oxidized. It is then transported to the Golgi apparatus where it is packaged into secretory vesicles, and where it is processed by a series of proteases to form mature insulin. Mature insulin has 35 fewer amino acids; 4 are removed altogether, and the remaining 31 form the C-peptide. The C-peptide is abstracted from the center of the proinsulin sequence; the two other ends (the B chain and A chain) remain connected by disulfide bonds. The post translational modification of proinsulin to mature insulin only occurs in the beta cells of the pancreatic islets. When proinsulin is transported through the Golgi apparatus the C-peptide is cleaved. This cleavage occurs with the aid of two endoproteases. Type I endoproteases, PC1 and PC3, disrupt the C peptide-B chain connection. PC2, a type II endoprotease, cleaves the C peptide-A chain bond. The resulting molecule, now mature insulin, is stored as a hexamer in secretory vesicles and is stabilized with
