Thymosin alpha-1 is a synthetic peptide of 28 amino acid residues that corresponds to a naturally occurring fragment first isolated from thymus tissue. Its chain is acetylated at the amino terminus, a modification that shields the peptide from rapid cleavage by aminopeptidases. The molecule carries a net negative charge at physiological pH and dissolves freely in water. Researchers classify it as an immune-modulating agent rather than a classical hormone, because it acts on several cell types of both the innate and the adaptive immune system.
The peptide was identified during work in the 1970s on thymosin fraction 5, a partially purified extract of calf thymus. Investigators separated that mixture and characterized individual components, one of which they named thymosin alpha-1. The same compound later received the international nonproprietary name thymalfasin. Commercial material is produced by solid-phase peptide synthesis rather than by extraction, so synthetic and natural forms share an identical sequence. Naming conventions vary across the literature, and readers should distinguish the alpha-1 peptide from other thymosins that have unrelated sequences and functions.

=== Permian reptiles === With the close of the Carboniferous, the amniotes became the dominant tetrapod fauna. While primitive, terrestrial reptiliomorphs still existed, the synapsid amniotes evolved the first truly terrestrial megafauna (giant animals) in the form of pelycosaurs, such as Edaphosaurus and the carnivorous Dimetrodon. In the mid-Permian period, the climate became drier, resulting in a change of fauna: The pelycosaurs were replaced by the therapsids. Many stem reptile groups continued to flourish throughout the Permian. The herbivorous pareiasaurs were the first lineage of reptiles to reach a large body size, with the largest representatives of the group having a body mass estimated to exceed 1,000 kilograms (2,200 lb). One of the best known early stem-reptiles is Mesosaurus, a genus from the Early Permian of Southern Africa and South America that had returned to water and gained webbed feet, feeding on crustaceans in marine or lagoonal environments. The earliest true diapsids (Neodiapsida), such as Youngina, appeared during the Middle-Late Permian. The Weigeltisauridae, a group of diapsids from the Late Permian, are the oldest known tetrapods to have engaged in flight, using novel rod-like bones extending from the trunk which formed wings to glide between trees.
== Regulation == IGFBP-3 levels in human serum are, like IGF-1, dependent on growth hormone (GH); for example, serum IGFBP-3 is increased in acromegaly and low in GH-deficient children. However, IGFBP-3 gene expression in human liver is GH-independent. Because it is stabilized in human serum by forming complexes with IGF-1 and ALS, which are both GH-dependent, serum IGFBP-3 also appears regulated by GH. Its production by some non-hepatic tissues may also be directly GH-regulated. Immunoassays for serum IGFBP-3 are often used as part of the diagnosis of childhood GH-deficiency. The most widely studied IGFBP3 polymorphism, at nucleotide-202 in the promoter region, is significantly associated with circulating IGFBP-3 levels, although the mechanism is unclear. In some studies circulating IGFBP-3 also appears to be nutritionally regulated, although this may not be seen at the mRNA level. IGFBP-3 has been identified in human lymph, nipple aspirate, milk, amniotic fluid, follicular fluid, seminal plasma, urine, peritoneal dialysate, synovial fluid, tear fluid, and cerebrospinal fluid, in addition to serum. Many factors increase IGFBP-3 production by cells, including transforming growth factor-β (TGFβ), tumor necrosis factor-α, vitamin D, retinoic acid, IGF-1, and stimuli such as chemotherapy that activate the tumor suppressor p53. Estrogen inhibits IGFBP-3 production, and its tissue levels are lower in estrogen receptor (ER)-positive breast cancers than in ER-negative cancers.
Hemangiopericytoma with t(7;12)(p22;q13)-translocations is a rare affliction, in which a translocational mutation causes the fusion of the ACTB gene over GLI1 in Chromosome 12. Juvenile onset dystonia is a rare degenerative disease that affects the central nervous system; in particular, it affects areas of the neocortex and thalamus, where rod-like eosinophilic inclusions are formed. The affected individuals represent a phenotype with deformities on the median line, sensory hearing loss and dystonia. It is caused by a point mutation in which the amino acid tryptophan replaces arginine in position 183. This alters actin's interaction with the ADF/cofilin system, which regulates the dynamics of nerve cell cytoskeleton formation. A dominant point mutation has also been discovered that causes neutrophil granulocyte dysfunction and recurring infections. It appears that the mutation modifies the domain responsible for binding between profilin and other regulatory proteins. Actin's affinity for profilin is greatly reduced in this allele. The ACTG1 locus codes for the cytosolic γ-actin protein that is responsible for the formation of cytoskeletal microfilaments. It contains six exons, giving rise to 22 different mRNAs, which produce four complete isoforms whose form of expression is probably dependent on the type of tissue they are found in. It also has two different DNA promoters.
In Latin America 62% of people believe that the level of corruption had risen in the last 12 months according to a study in 2017. People feel this way due to a number of ways in which governments use corruption to gain money and power at the expense of their people. In Venezuela the Maduro regime has been known to allow transnational criminal organizations. This is along with money laundering, or illicit finance, which keeps money in the pockets of criminals and government officials instead of the people.

== Medical uses == Prazosin is active after taken by mouth and has a minimal effect on cardiac function due to its α1-adrenergic receptor selectivity. When prazosin is started, however, heart rate and contractility can increase in order to maintain the pre-treatment blood pressures because the body has reached homeostasis at its abnormally high blood pressure. The blood pressure lowering effect becomes apparent when prazosin is taken for longer periods of time. The heart rate and contractility go back down over time and blood pressure decreases. The antihypertensive characteristics of prazosin make it a second-line choice for the treatment of high blood pressure. Prazosin is also useful in treating urinary hesitancy associated with benign prostatic hyperplasia, blocking α1-adrenergic receptors, which control constriction of both the prostate and urethra. Although not a first-line choice for either hypertension or benign prostatic hyperplasia, it is a choice for people who present with both problems concomitantly. During its use for urinary hesitancy in military veterans in the 1990s, Murray A. Raskind and colleagues discovered that prazosin appeared to be effective in reducing nightmares. Subsequent reviews indicate prazosin is effective in improving sleep quality and treating nightmares related to post-traumatic stress disorder (PTSD). Prazosin is used off-label in the treatment of insomnia for its sedative effects. Prazosin is an inverse agonist at α1-adrenergic receptors; these receptors are expressed on dendrites that noradrenergic neurons synapse onto in the brain.
=== Pharmacokinetics === In non-human primates, the plasma half-life of the prodrug is 20 minutes, with the main metabolite being the nucleoside, GS-441524. Two hours post injection, the main metabolite GS-441524 is present at micromolar concentrations, whilst intact Remdesivir is no longer detectable. Because of this rapid extracellular conversion to the nucleoside GS-441524, some researchers have questioned whether the active nucleotide triphosphate is truly derived from remdesivir pro-drug removal or whether it occurs by GS-441524 phosphorylation, and whether direct administration of GS-441524 would constitute a cheaper and easier to administer COVID‑19 drug compared to remdesivir. The activated nucleotide triphosphate form has sustained intracellular levels in PBMC and presumably in other cells as well.
The branched-chain fatty acid synthesizing system uses α-keto acids as primers. This system is distinct from the branched-chain fatty acid synthetase that utilizes short-chain acyl-CoA esters as primers. α-Keto acid primers are derived from the transamination and decarboxylation of valine, leucine, and isoleucine to form 2-methylpropanyl-CoA, 3-methylbutyryl-CoA, and 2-methylbutyryl-CoA, respectively. 2-Methylpropanyl-CoA primers derived from valine are elongated to produce even-numbered iso-series fatty acids such as 14-methyl-pentadecanoic (isopalmitic) acid, and 3-methylbutyryl-CoA primers from leucine may be used to form odd-numbered iso-series fatty acids such as 13-methyl-tetradecanoic acid. 2-Methylbutyryl-CoA primers from isoleucine are elongated to form anteiso-series fatty acids containing an odd number of carbon atoms such as 12-Methyl tetradecanoic acid. Decarboxylation of the primer precursors occurs through the branched-chain α-keto acid decarboxylase (BCKA) enzyme. Elongation of the fatty acid follows the same biosynthetic pathway in Escherichia coli used to produce straight-chain fatty acids where malonyl-CoA is used as a chain extender. The major end products are 12–17 carbon branched-chain fatty acids and their composition tends to be uniform and characteristic for many bacterial species. BCKA decarboxylase and relative activities of α-keto acid substrates The BCKA decarboxylase enzyme is composed of two subunits in a tetrameric structure (A2B2) and is essential for the synthesis of branched-chain fatty acids.
== Development of the United Nations == Although the United States never joined the League of Nations, the Rockefeller Foundation was involved, and by the 1930s the foundations had changed the League from a "Parliament of Nations" to a modern think tank that used specialized expertise to provide in-depth impartial analysis of international issues. After the war, the foundation was involved in the establishment of the United Nations.
molecular biology The branch of biology that studies biological activity at the molecular level, in particular the various mechanisms underlying the biological processes that occur in and between cells, including the structures, properties, synthesis, and modification of biomolecules such as proteins and nucleic acids, their interactions with the chemical environment and with other biomolecules, and how these interactions explain the observations of classical biology (which in contrast studies biological systems at much larger scales). Molecular biology relies largely on laboratory techniques of physics and chemistry to manipulate and measure microscopic phenomena. It is closely related to and overlaps with the fields of cell biology, biochemistry, and molecular genetics.

It is a 28-residue synthetic peptide studied as an immune-modulating agent and approved as a drug in some countries. The sequence matches a naturally occurring fragment isolated from thymus tissue. It is not a hormone in the endocrine sense.
The name traces back to thymosin fraction 5, a crude thymus extract examined in the 1970s. Individual peptides in that mixture were labeled with Greek letters, and alpha-1 was one of them. The international nonproprietary name thymalfasin was assigned later.
No. Thymosin beta-4 contains 43 residues and binds actin, while thymosin alpha-1 contains 28 residues and acts on immune cells. The two sit in a historical naming group but share no sequence similarity, and they are not substitutes for one another.
