Lyophilized thymosin alpha-1 is generally stored at or below minus twenty degrees Celsius, protected from moisture and light. Short-term handling at ambient temperature is possible for dry powder, but reconstituted solutions degrade faster and are usually kept at two to eight degrees Celsius with a defined expiry of days rather than weeks. Repeated freeze-thaw cycles should be avoided because they promote aggregation and loss of potency. Exact limits depend on the formulation and should follow the supplier's documentation.
Identity and purity are normally assessed by reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities and truncation products. Mass spectrometry confirms molecular mass and detects modifications such as deamidation or oxidation. Amino acid analysis and peptide mapping provide additional sequence-level confirmation. For research material, a certificate of analysis typically reports these results together with water content and counter-ion identity, since the lyophilized powder is often supplied as an acetate or trifluoroacetate salt.
Lyophilized material is typically treated as a hygroscopic solid that should be brought to room temperature before the container is opened, which limits condensation on the powder. Reconstitution is commonly done with sterile water or a buffered diluent, and gentle mixing is preferred over vigorous agitation to reduce foaming and surface adsorption. Because peptides can bind to plastic and glass, diluents containing a small amount of carrier protein are sometimes used in laboratory work. Working solutions are generally aliquoted and prepared fresh rather than subjected to repeated freezing and thawing.
Stability depends on temperature, pH, and the number of freeze-thaw events the sample has experienced. Freeze-dried material is commonly held at -20 °C or colder, while reconstituted liquid is kept cold and used within a short window. Extreme pH and prolonged light exposure can promote deamidation, oxidation, or aggregation, particularly at asparagine and methionine positions. Adsorption to container walls can lower the measured concentration of a dilute solution even when the peptide molecules themselves remain intact.

Thymosin alpha-1 is a synthetic peptide of 28 amino acids, corresponding to the N-terminal fragment of prothymosin alpha. Its sequence begins with acetylation at the N-terminus, a modification that affects stability and receptor interaction. The peptide is acidic, with a calculated isoelectric point near 4.2, and carries no disulfide bonds, so its secondary structure is largely flexible in solution. Molecular mass is approximately 3108 daltons. The native form was first isolated from bovine thymus tissue, while pharmaceutical material is produced by solid-phase peptide synthesis.
Within the immune system, the peptide acts on several cell types rather than a single target. Reported activities include promotion of T-cell maturation, enhancement of natural killer cell activity, and modulation of cytokine production by dendritic cells and macrophages. Some of these effects appear to operate through toll-like receptor signaling, though the precise receptor-level mechanism remains debated. Whether the observed immune changes translate into clinical benefit is a separate question and depends on the indication studied.
The peptide was described in the 1970s as a component of thymic extracts, and early research focused on restoring immune function in immunodeficiency states. A synthetic version entered clinical development in the 1980s and is approved as a drug in several countries for conditions such as chronic hepatitis B and certain immunodeficiencies. Approval status varies widely by jurisdiction, and in the United States it is not an approved therapeutic. Regulatory and clinical positions differ, so statements about efficacy should be tied to specific indications and studies.
== Early political career (1959–1970) == After World War II ended and the Japanese withdrew, the British grouped the Malay states and the Straits Settlements into the Malayan Union, and granted citizenship to non-Malays. This caused major backlash from Malays and a wave of Malay nationalism swept across the country. Mahathir became politically activated by these changes, joining protests and activism against the new citizenship policies. Mahathir later argued for affirmative action for Malays at medical college. While at college, he contributed to The Straits Times under the pseudonym "C.H.E. Det" and a student journal, in which he fiercely promoted Malay rights, such as calling for the restoration of Malay as an official language. While practising as a physician in Alor Setar, Mahathir became active in UMNO. His earliest political involvement was in the Kampung Charok Kudong Umno branch, where he made his debut in politics. By the time of the first general election for the independent state of Malaya in 1959, he was the chairman of the party in Kedah. Despite his prominence in UMNO, Mahathir was not a candidate in the 1959 election, ruling himself out following a disagreement with then Prime Minister Tunku Abdul Rahman. Their relationship had been strained since Mahathir had criticised Tunku's agreement to retain British and Commonwealth forces in Malaya after independence. Tunku opposed Mahathir's plans to introduce minimum educational qualifications for UMNO candidates.
The perianth is undifferentiated (perigonium) and biseriate (two whorled), formed from six tepals arranged into two separate whorls of three parts (trimerous) each, although Scoliopus has only three petals, free from the other parts, but overlapping. The tepals are usually petaloid (petal like) and apotepalous (free) with lines (striate) or marks in other colors or shades. The perianth is either homochlamydeous (all tepals equal, e.g. Fritillaria) or dichlamydeous (two separate and different whorls, e.g. Calochortus) and may be united into a tube. Nectar is produced in perigonal nectaries at the base of the tepals. The androecium consists of six stamens in two trimerous whorls, with free filaments, usually epiphyllous (fused to tepals) and diplostemonous (outer whorl of stamens opposite outer tepals and the inner whorl opposite inner tepals), although Scoliopus has three stamens opposite the outer tepals. The attachment of the anthers to the filaments may be either peltate (to the surface) or pseudo-basifixed (surrounding the filament tip, but not adnate, that is not fused) and dehisce longitudinally and are extrorse (dehiscing away from center). The pollen is usually monosulcate (single groove), but may be inaperturate (lacking aperture: Clintonia, some Tulipa spp.) or operculate (lidded: Fritillaria, some Tulipa spp.), and reticulate (net patterned: Erythronium, Fritillaria, Gagea, Lilium, Tulipa).
Duchenne muscular dystrophy is caused by a mutation of the dystrophin gene, located on the short arm of the X chromosome (locus Xp21) that codes for dystrophin protein. Mutations can either be inherited or occur spontaneously during germline transmission, causing a large reduction or absence of dystrophin, a protein that provides structural integrity in muscle cells. Dystrophin is responsible for connecting the actin cytoskeleton of each muscle fiber to the underlying basal lamina (extracellular matrix), through a protein complex containing many subunits. The absence of dystrophin permits excess calcium to penetrate the sarcolemma (the muscle cell membrane).

Calcitonin gene-related peptide (CGRP) receptor antagonists, commonly known as gepants, are a class of drugs that act as antagonists of the calcitonin gene-related peptide receptor (CGRPR). The CGRP family of small proteins are present in the sensory nerves of the head and neck and are involved in transmission of pain. Nerve activation can trigger the release of CGRP and other neuropeptides, leading to inflammation, pain, and swelling in the case of migraine. Several monoclonal antibodies that bind to the CGRP receptor or peptide have been approved for prevention of migraine. As of March 11, 2024, the American Headache Society issued a statement that "CGRP targeting therapies are a first-line option for migraine prevention" in the United States. The prior use of non-specific migraine preventive medication approaches is therefore no longer required before CGRP treatments can be prescribed. Small molecule CGRPR antagonists have also been approved in the U.S. as antimigraine agents.
== Function == Mambalgins are potent inhibitors of acid-sensing ion channels (ASICs), which are multimeric membrane proteins that respond to low pH and whose activation is thought to be involved in perception of pain. Mambalgins have been shown to interact specifically with ASIC subtypes present in the central nervous system (homomeric ASIC1a and heteromeric ASIC1a/ASIC2a or ASIC1a/ASIC2b), as well as those found in sensory neurons (ASIC1b and ASIC1a/ASIC1b). They have no effect on other ASICs or on other types of ion channel proteins. These interactions are likely mediated in part by mambalgins' positive electrostatic potential facilitating binding to negatively charged ASICs. Mambalgins are believed to trap ASICs in a closed conformation. In tests performed on laboratory mice, mambalgins have the in vivo effect of analgesia without the toxic effects seen with most 3FTx proteins, and in particular, without the clinical manifestations associated with inhibition of nicotinic acetylcholine receptors, the targets of most 3FTx proteins including mambalgins' closest relatives. Furthermore, the analgesic effects of mambalgins does not confer side effects such as respiratory depression and drug tolerance, both associated with opioid analgesics.
=== Flavour and aroma === Dozens of compounds contribute to Scotch whisky flavour and aroma characteristics, including volatile alcohol congeners (also called higher oils) formed during fermentation, such as acetaldehyde, methanol, ethyl acetate, n-propanol and isobutanol. Other flavour and aroma compounds include vanillic acid, syringic acid, vanillin, syringaldehyde, furfural, phenyl ethanol and acetic acid. One analysis established 13 distinct flavour characteristics dependent on individual compounds, including sour, sweet, grainy and floral as major flavour perceptions. Some distilleries use a peat fire to dry the barley for some of their products before grinding it and making the mash. Peat smoke contributes phenolic compounds, such as guaiacol, that give aromas similar to smoke. The Maillard browning process of the residual sugars in the mashing process, particularly through formation of 2-furanmethanol and pyrazines imparting nutty or cereal characteristics contributes to the baked bread notes in the flavour and aroma profile. Maturation during multi-year casking in oak barrels mostly previously used for bourbon whiskey, sherry, wines, fortified wine (including port and Madeira), rum and other spirit production, has the largest impact on the flavour of the whisky. Some distilleries use virgin oak casks as used casks are becoming increasingly harder to source (particularly authentic sherry casks due to the downturn in sherry consumption plus the laws introduced in 1986 regarding bottling Spanish wines exclusively in Spain).
== Side effects == Protriptyline shares side effects common to all TCAs. The most frequent of these are dry mouth, constipation, urinary retention, increased heart rate, sedation, irritability, decreased coordination, anxiety, blood disorders, confusion, decreased libido, dizziness, flushing, headache, impotence, insomnia, low blood pressure, nightmares, rapid or irregular heartbeat, rash, seizures, sensitivity to sunlight, stomach and intestinal problems. Other more complicated side effects include; chest pain or heavy feeling, pain spreading to the arm or shoulder, nausea, sweating, general ill feeling; sudden numbness or weakness, especially on one side of the body; sudden headache, confusion, problems with vision, speech, or balance; hallucinations, or seizure (convulsions); easy bruising or bleeding, unusual weakness; restless muscle movements in your eyes, tongue, jaw, or neck; urinating less than usual or not at all; extreme thirst with headache, nausea, vomiting, and weakness; or feeling light-headed or fainting. Dry mouth, if severe to the point of causing difficulty speaking or swallowing, may be managed by dosage reduction or temporary discontinuation of the drug. Patients may also chew sugarless gum or suck on sugarless candy in order to increase the flow of saliva. Some artificial saliva products may give temporary relief. Men with prostate enlargement who take protriptyline may be especially likely to have problems with urinary retention. Symptoms include having difficulty starting a urine flow and more difficulty than usual passing urine.

=== General mixtures === Insulin mixtures can sometimes be combined in a single syringe, such as when mixing intermediate-acting insulin with rapid- or short-acting insulin. However, not all types of insulin are compatible for mixing. For convenience, there are premixed formulations of rapid- and intermediate-acting insulins. These come in fixed ratios like 75/25, 70/30, or 50/50. For example, a 75/25 mix contains 75% intermediate-acting insulin and 25% rapid-acting insulin. These are typically injected twice a day at the start of meals. The mixture appears cloudy, and it begins to work as quickly as the rapid-acting insulin, but it lasts as long as the intermediate-acting insulin.
==== History ==== It was founded in March 1979 as part of the People's Liberation Army Basic Engineering Corps. It was under the joint dual command of the Basic Engineering Corps and the Ministry of Metallurgical Industry. It consisted of 7 regiment level battalions and 12 geological teams. The Gold Corps was handed to the PAP in January 1985. After 1985, it went under the dual control of the Ministry of Metallurgical Industry and Ministry of Public Security. Its role in this time was to do geological surveys for gold mining. After February 1999, it went under the dual control of the People's Armed Police and the State Economic and Trade Commission. Its official duties were: finding gold deposits, providing security and law enforcement to gold mines, mining gold and preventing conflict near gold mines. By 2011, the gold corps had mined 1800 tonnes of gold. The same year in December, its official role switched again to geological surveying and metal analysis along with disaster relief to geological related incidents and law enforcement. In 2016, its role was switched yet again to focus primarily on military geology related work in the Deepening National Defense and Military Reform. In 2018, the Gold Corps was disbanded and its personnel were handed to the Ministry of Natural Resources, the China Geological Survey and the China National Gold Group Corporation.
== Signs and symptoms == The characteristics of a burn depend upon its depth. Superficial burns cause pain lasting two or three days, followed by peeling of the skin over the next few days. Individuals with more severe burns may indicate discomfort or complain of feeling pressure rather than pain. Full-thickness burns may be entirely insensitive to light touch or puncture. While superficial burns are typically red in color, severe burns may be pink, white or black. Burns around the mouth or singed hair inside the nose may indicate that burns to the airways have occurred, but these findings are not definitive. More worrisome signs include: shortness of breath, hoarseness, and stridor or wheezing. Itchiness is common during the healing process, occurring in up to 90% of adults and nearly all children. Numbness or tingling may persist for a prolonged period of time after an electrical injury. Burns may also produce emotional and psychological distress.

Dry lyophilized powder tolerates short ambient exposure during handling and shipping. Long-term room-temperature storage is not recommended because moisture uptake and slow degradation can occur over months. Storage at minus twenty degrees Celsius is the common practice for extended periods.
Typical entries list appearance, identity by mass spectrometry, purity by chromatography, water or moisture content, and residual counter-ion. Some certificates also report microbial limits and endotoxin for materials intended for laboratory use. The reported methods and acceptance ranges vary between suppliers.
The peptide is often supplied as an acetate or trifluoroacetate salt, and the counter-ion affects solubility, weight-per-mole calculations, and compatibility with cell assays. Trifluoroacetate can be undesirable in some biological experiments. Knowing the salt form is necessary for accurate concentration determination.
Cold storage slows the chemical degradation reactions that occur in solution. Lyophilized powder is more stable than reconstituted liquid and tolerates longer storage periods. Repeated temperature cycling should still be avoided because it can drive aggregation and loss of material.
